Mammalian cell encapsulation in monodisperse chitosan beads using microchannel emulsification

Microchannel emulsification can produce droplets of uniform size at high throughput for wide-ranging applications in the food and drug industry. Mammalian cell encapsulation using emulsion-based systems is significant interest immobilized culture or transplantation applications. has recently been applied to mammalian alginate. Here, we extend this approach thermoresponsive hydrogels chitosan as a model. The performance microchannel process was compared previously established stirred method mesenchymal stromal encapsulation. Chitosan microbeads with controlled sizes ranging from 600 µm 1500 diameter coefficient variation less than 10 % were obtained after adapting system design, channel geometries operating parameters that affect capillary number. In single pass through 21 G syringe needle, fraction ruptured beads significantly reduced monodisperse microchannel-generated vs polydisperse emulsion-generated matching De Brouckere mean diameter. viability cells immediately 95 ± 2 no difference survival growth factor secretion observed between over 3 days culture. promising scalable microencapsulate undergo temperature-dependent gelation.
